RED CELLS Species- and cell type–specific interactions between CD47 and human SIRP

چکیده
CD47 on red blood cells (RBCs) reportedly signals “self” by binding SIRP on phagocytes, at least in mice. Such interactions across and within species, from mouse to human, are not yet clear and neither is the relation to cell adhesion. Using human SIRP 1 as a probe, antibody-inhibitable binding to CD47 was found only with human and pig RBCs (not mouse, rat, or cow). In addition, CD47mediated adhesion of human and pig RBCs to SIRP 1 surfaces resists sustained forces in centrifugation (as confirmed by atomic force microscopy) but only at SIRP -coating densities far above those measurable on human neutrophils, monocytes, and THP-1 macrophages. While interactions strengthen with deglycosylation of SIRP 1, low copy numbers explain the absence of RBC adhesion to phagocytes under physiologic conditions and imply that the interaction being studied is not responsible for red cell clearance in humans. Evidence of clustering nonetheless suggests mechanisms of avidity enhancement. Finally, using the same CD47 antibodies and soluble SIRP 1, bone marrow–derived mesenchymal stem cells were assayed and found to display CD47 but not bind SIRP 1 significantly. The results thus demonstrate that SIRP -CD47 interactions, which reportedly define self, exhibit cell type specificity and limited cross-species reactivity. (Blood. 2006;107:2548-2556)

CD47Low Status on CD4 Effectors Is Necessary for the Contraction/Resolution of the Immune Response in Humans and Mice
How do effector CD4 T cells escape cell death during the contraction of the immune response (IR) remain largely unknown. CD47, through interactions with thrombospondin-1 (TSP-1) and SIRP-α, is implicated in cell death and phagocytosis of malignant cells. Here, we reported a reduction in SIRP-α-Fc binding to effector memory T cells (T(EM)) and in vitro TCR-activated human CD4 T cells that was li...

Signal-regulatory proteins (SIRPs) are transmembrane glycoproteins belonging to the immunoglobulin (Ig) superfamily that are expressed in the immune and central nervous systems. SIRP binds CD47 and inhibits the function of macrophages, dendritic cells, and granulocytes, whereas SIRP 1 is an orphan receptor that activates the same cell types. A recently identified third member of the SIRP family...

متن کاملRED CELLS, IRON, AND ERYTHROPOIESIS CD47 functions as a molecular switch for erythrocyte phagocytosis
CD47 on erythrocytes inhibits phagocytosis through interaction with the inhibitory immunoreceptor SIRP expressed by macrophages. Thus, the CD47-SIRP interaction constitutes a negative signal for erythrocyte phagocytosis. However, we report here that CD47 does not only function as a “do not eat me” signal for uptake but can also act as an “eat me” signal. In particular, a subset of old erythrocy...
